چکیده

BACKGROUND Peripheral blood leukocytes (PBLs) are the main source of DNA in blood samples. A common protocol is to store buffy coat specimens for future DNA isolation, and such samples may remain in frozen storage for years. However, the currently available methods to maintain buffy coat specimens can be optimized for better quality and cost efficiency. STUDY DESIGN AND METHODS Seventeen donors (aged 24-34 years) were enrolled in this study. Initially, five centrifugal speeds were chosen to examine the distribution of PBLs in the cell layer; the buffy coat was then harvested to evaluate the cell quantity and viability after storing at 4°C for various times. Finally, the buffy coat was isolated, snap frozen, and kept at -80°C for 1 hour, 1 week, or 4 weeks until the DNA was extracted. Agarose gel electrophoresis and multiplex PCR were used to verify DNA integrity and amplifiability. RESULTS There was a linear positive correlation between the amount of fresh PBLs and the DNA yield. At least 70% of PBLs were collected in the uppermost 40% of the cellular material when the centrifugal force was over 800 g compared to 400 g. Storing blood at 4°C for no more than 24 hours did not have an effect on the amount of PBLs or their viability. In addition, the amount of extracted DNA was decreased in the frozen buffy coat that was stored for more than 7 days, though the DNA quality was acceptable. CONCLUSION DNA should be extracted from fresh buffy coat samples as soon as possible after collection, especially for very important samples. Retaining the upper 40% of the cell pack of blood instead of whole blood could improve the storage efficiency of biobanking such samples. Our study provides a recommended option for blood collection and processing protocols in biobanking.

Effect of famotidine on radiation induced apoptosis in human peripheral blood leukocytes

ABSTRACTBackground: Radioprotective effects of famotidine, an antagonist of H2 receptor clinically used for peptic ulcer treatment, was previously shown on radiation-induced micronuclei and chromosomal aberration in human peripheral blood lymphocytes and mouse bone marrow cells. This study was conducted to investigate radioprotective property of famotidine against radiation induced apoptosis i...

Radioadaptive response in peripheral blood leukocytes of occupationally exposed medical staff with investigation of DNA damage by the use of neutral comet assay

Background: ˝Radioadaptive Response˝ is well-documented phenomenon appeared in low dose ionizing radiation received in vitro and in vivo. Occupational exposure has always been a great concern for radiation workers therefore this study was performed to study radioadaptive response in terms of residual DNA double strand breaks as an endpoint in peripheral blood leukocytes of occupationally expose...

Consumption of antioxidant dietary agents, curcumin and vitamin C, protects cellular DNA from gamma-radiation

Background: Exposure to ionizing radiation results in genotoxicity and the unrepaired lesions in cellular DNA results in cell cycle arrest, reproductive death, interphase death, division delay, chromosome aberrations, mutations, etc. leading to the intensive destruction of cells and violation of their proliferative capacity there by adversely affecting the mammalian system. Since ionizing radia...

Assessment of mitochondrial DNA copy number in peripheral blood leukocyte of opiate abusers and healthy individuals

Background: Based on the studies, variation in the mitochondrial DNA (mtDNA) copy number in peripheral blood leukocytes is associated with increased susceptibility to diseases including cancer. Opiate abusers are at high risk for diseases. In this study, we measured the mtDNA copy number in peripheral blood leukocytes in a group of opiate abusers compared with those in healthy individuals. Met...

Interindividual differences in radiation-induced apoptosis of peripheral blood leukocytes in normal individuals and breast cancer patients

Background: Quantification of radiation-induced apoptosis in peripheral blood lymphocytes (PBLs) has been proposed as a possible screening test for cancer-prone individuals and also for the prediction of normal tissue responses after radiotherapy. Materials and Methods: The neutral version of the comet assay (single-cell gel electrophoresis) was used 24, 48, 72 hours after irradiation ...
